چکیده

In addition to its effect on Trichomonas vaginalis, metronidazole has also been found to be active against the organisms causing Vincent's gingivitis. This has raised the question of its possible action on Treponema pallidum, although Scott-Gray and Murrell (1961) found that a short course of 1-8 g. given over 48 hours did not affect a case of primary syphilis, and Yobs, Clark, and Schroeter (1966) found that large doses did not affect the course of syphilis in rabbits infected with the Nichols strain of Trepomena pallidum. Davies (1967), however, has reported that doses of 2 to 4 g. daily caused the disappearance of Treponema pallidum from the lesions of six patients with secondary syphilis with subsequent healing of the lesions; smaller doses were without effect. Davies, McFadzean, and Squires (1964) have reported that the minimum inhibitory concentration of metronidazole for the cultivable Reiter treponeme is 0 02 [±g./ml. These observations prompted the examination of the effect of serum containing metronidazole on the Nichols strain of Treponema pallidum in vitro.

In vitro culture system to determine MICs and MBCs of antimicrobial agents against Treponema pallidum subsp. pallidum (Nichols strain).

A new procedure for determining the susceptibility of Treponema pallidum subsp. pallidum to antimicrobial agents was developed, utilizing a tissue culture system which promotes the in vitro multiplication of this organism. In the absence of antibiotics, T. pallidum (Nichols virulent strain) multiplied an average of 10-fold when incubated for 7 days in the presence of Sf1Ep cottontail rabbit epi...

TprK sequence diversity accumulates during infection of rabbits with Treponema pallidum subsp. pallidum Nichols strain.

The tprK gene in Treponema pallidum undergoes antigenic variation. In all T. pallidum isolates examined to date, except the Nichols type strain, heterogeneous tprK sequences have been identified. This heterogeneity is localized to seven variable (V) regions, and tprK sequence diversity accumulates with serial passage in naïve rabbits. The T. pallidum Nichols genome described a single tprK seque...

Ultrastructural studies of treponemes: location of axial filaments and some dimensions of Treponema pallidum (Nichols strain), Treponema denticola, and Treponema reiteri.

Ultrathin sections of Treponema pallidum (Nichols strain), T. denticola (microdentium), and T. reiteri have been studied in the electron microscope to determine the location of the axial filaments and some of the dimensions of these organisms. The axial filaments of T. pallidum (Nichols strain) have been seen to be tubular in cross section with an overall diameter of 21.0 +/- 0.73 nm, and an el...

Complete genome sequence of Treponema pallidum strain DAL-1

Treponema pallidum strain DAL-1 is a human uncultivable pathogen causing the sexually transmitted disease syphilis. Strain DAL-1 was isolated from the amniotic fluid of a pregnant woman in the secondary stage of syphilis. Here we describe the 1,139,971 bp long genome of T. pallidum strain DAL-1 which was sequenced using two independent sequencing methods (454 pyrosequencing and Illumina). In ra...

Genetic relationship between Treponema pallidum and Treponema pertenue, two noncultivable human pathogens.

Genetic relationships among two strains of Treponema pallidum (Nichols and KKJ) and a strain of T. pertenue were determined by measuring the degree of deoxyribonucleic acid sequence homology. The results in indicated that these three virulent, noncultivable treponemes were genetically indistinguishable. Like T. pallidum (Nichols), T. pertenue (Gauthier) had no detectable deoxyribonucleic acid s...
